Regulation of actin and microtubule cytoskeleton by MAP6


The aim of this study is to dissect the mode of action of MAP6 on actin and microtubules dynamics and structure through the identification of the domains of the protein involved in these different functions.


MAP6 plays a role in neuronal development, signaling and plasticity. MAP6-KO mice show behavioral alterations reminiscent of schizophrenia and are used as animal model for this neuropathology. MAP6 interacts in vivo with microtubules and actin filaments, the two main cytoskeleton components that are essential for neuronal development and functions. We have found that in vitro MAP6 regulates microtubule nucleation, dynamics and structure. MAP6 also promotes actin filament polymerization and bundling. The project aims at a better understanding of the mode of action of MAP6 by the analysis of the effects of purified recombinant fragments of the MAP6 protein on actin and microtubules dynamics and structure using biochemical, TIRF microscopy and electron microscopy approaches.
