Deciphering the interaction of the neuronal MAP6 with microtubules


The objective of this study is to dissect the molecular MAP6 (Microtubule-Associated Protein 6) domains involved in the interaction and regulation of microtubules dynamics and structure.


MAP6 plays a role in the development, signaling and plasticity of neurons. MAP6-KO mice exhibit behavioural alterations reminiscent of schizophrenia and are used as an animal model for this neuropathology. These behavioural alterations are partly due to the ability of MAP6 to regulate microtubule dynamics. A recent study by the team shows that MAP6 binds to both the outer and inner side of the tube. The presence of proteins inside neural microtubules and their physiological roles has been a long-standing question. The identification of MAP6 as the first neural microtubule internal protein (MIP) gives us the tools to study the role of neural MIPs. To understand the neuronal function of the association of MAP6 with the inner or outer side of the microtubule, specific microtubule binding mutants are needed and must be characterized. The project consists of analyzing the effects of MAP6 mutant recombinant proteins on the dynamics and structure of microtubules using biochemical, TIRF and electron microscopy approaches.
